Top button with no tie?
I feels it’s generally acceptable to leave open the top button. It’s more comfortable, and creates a more relaxed vibe. Sure, if what you’re aiming for is a more professional look then go ahead and button up.
Polos aren’t really formal (my unpopular opinion)
The only acceptable time to button the top button on any shirt is if you are also wearing a tie. Otherwise you just look like Dr Evil.
Undershirt: Unbuttoned.
No Undershirt: Buttoned.
It’s a polo shirt, it’s casual, why do you want to keep buttons done up to look formal? If you want formal wear a proper shirt.

Nothing about a polo shirt looks formal. It’s not supposed to look formal.
Buttoning the top button of a shirt when you’re not wearing a tie both looks ridiculous and makes you look needlessly uncomfortable.
If it was up to me I wouldn’t be wearing slacks and a polo shirt to work but people are still caught up on what you wear is what makes you a professional. So I’m going to leave the top button undone cause I’m already uncomfortable af
We have polo shirts at our company, and there the unspoken rule is to button two of three buttons. Only one looks too lax. Buttoning ’em all looks too stiff.

Hot take, but having the polo completely buttoned makes you look like a nerd. You might as well get some rainbow suspenders hand wear your trousers up by your pecks and wear Harry Potter goggles with a white bandage around the nose-brim.

Etiquette is, that if you’re not wearing a tie, you don’t fully button your dress shirt or your polo shirt.
